The fixing screw can corrode and seize so this may be what you are looking at.

It is possible the clothing may get damaged if the cover is not in place but it is not likely to damage the machine.

It would depend on if the cover is damaged, the screw is likely to be fitted with a thread locking paint to prevent it coming loose. Use a good fitting 'quality' screwdriver and 'shock' the screw by hitting the screwdriver handle to punch the tip tightly into the screw before trying to turn it. You may need an impact driver if this does not work.

If you cannot remove the screw inside the drum, is there another solution or another part that can be replaced? Any advice would be greatly appreciated?

It may be necessary to drill out the old screw and use a screw extractor to remove the stub but if this does not work you may need to replace the drum.
